A recent paper in Nature Communications titled ‘Histone methyltransferase PRDM9 promotes survival of drug-tolerant persister cells in glioblastoma’ by G.L. Joun and L. Munoz et al, uncovers a critical mechanism of chemotherapy resistance.
Chemotherapy often leaves behind drug-tolerant persister cells that drive recurrence. This study shows that PRDM9, a germ-cell-specific H3K4 methyltransferase, enables these persisters to survive by:
- Rewiring metabolism through enhanced cholesterol biosynthesis
- Helping cells withstand oxidative stress and lipid peroxidation
Importantly, PRDM9 inhibition combined with chemotherapy significantly improves anti-tumor efficacy in preclinical GBM models by eliminating persisters.
