”Our new paper has just been published in Nature Biomedical Engineering!
How can we make CAR-T therapy more durable and effective?
CAR-T cell therapy has changed the lives of many patients with blood cancers. However, one big challenge remains: many patients relapse because the CAR-T cells don’t last long enough or stop working over time.
In our study, we developed a vaccine-like approach that gives CAR-T cells a “boost” inside the body. By using specially designed synthetic peptides (mimotopes), we were able to help CAR-T cells expand, persist, and fight cancer more effectively.
Key highlights:
- We developed a general strategy to identify and optimize mimotopes for any CAR using yeast surface display + directed evolution.
- We demonstrated that CAR-T vaccines significantly boost CAR-T expansion, persistence, and memory in both mouse and humanized leukemia models.
- Achieved better leukemia control compared to CAR-T therapy alone.
This approach opens the door to applying vaccine boosting across diverse CAR-T products, offering hope for more effective and long-lasting cancer therapies.”
