Lei Deng: Could Lorlatinib Challenge the Belief That TKIs Cannot Cure ALK+ Lung Cancer?

Lei Deng, Thoracic Oncologist, Assistant Professor at University of Washington and Fred Hutc, shared a post by Aakash Desai, Associate Director, Phase 1 and Precision Oncology Program at UAB O’Neal Comprehensive Cancer Center, adding:

“With the pCR data (46.9% in resected; 35% ITT) of neoadjuvant lorlatinib (LORIN) and CROWN’s 7 year PFS rate of 55%, I wonder if we should reconsider whether the long-held belief that TKI cannot cure remains true in ALK.

Quoting Aakash Desai’s post:

CROWN at 7 years. Lorlatinib median PFS: still not reached.

7-yr PFS: 55% vs 3%. HR 0.19.
 Event-free at 2 years – 79% still progression-free at year 7.
 Zero new brain metastases after month 30. IC HR 0.06. Only 5% discontinued for TRAEs.

I think this is what it is for a meaningful subset of ALK+ patients: functional cure. Really exciting! For me today most patients should be getting lorlatinib 1L for ALK-Positive lung cancer.”
