The PADMA (GBG93) study is the first randomized multicenter trial to demonstrate that palbociclib combined with endocrine therapy (ET) is superior to standard single-agent chemotherapy as first-line treatment for patients with HR-positive/HER2-negative metastatic breast cancer, even in a high-risk population with a clinical indication for chemotherapy.
Key findings
- Time-to-treatment failure (TTF), the primary endpoint, was nearly tripled with palbociclib + ET:
17.2 months vs. 6.1 months with chemotherapy
Corresponding to a 54% reduction in risk. - Progression-free survival (PFS) was also significantly prolonged:
18.7 months vs. 7.8 months
Corresponding to a 55% reduction in risk. - The benefit was consistent across all analyzed subgroups, including patients with AKT pathway alterations or BRCA1/2 mutations.”
