“ESMO 2026 | ARTEMIS-011: first progress in decades for relapsed or refractory osteosarcoma with an antibody-drug conjugate.
Treatment options for relapsed or refractory osteosarcoma are extremely limited, with little progress since the introduction of chemotherapy. The phase III ARTEMIS-011 trial, presented at the Presidential Symposium (LBA9), tested the B7-H3-directed ADC risvutatug rezetecan against gemcitabine plus docetaxel in 122 adolescent and adult patients.
- Median progression-free survival by blinded independent central review (primary endpoint): 5.5 vs 1.4 months (HR 0.33; 95% CI 0.20–0.55; p<0.0001)
- Overall response rate with the ADC alone: 15%
- Overall survival is immature, with a favourable trend (HR 0.51; 95% CI 0.21–1.23)
- The safety profile was manageable, with one case of treatment-related interstitial lung disease
Commenting for ESMO, Dr. Nathalie Gaspar noted the limits: the biology behind the ADC’s efficacy is not yet fully understood, and haematological toxicity could be a problem alongside chemotherapy. Open questions include which target and payload work best, whether ADCs should move into earlier lines of treatment, and whether to combine them with chemotherapy, multikinase inhibitors or immunotherapy.”
