Abdu Adem Yesufe, Program Director of Adult Hematology and Medical Oncology Fellowship at St. Paul’s Hospital Millennium Medical College (SPHMMC), shared a post by Tesfaye Belete, Medical Doctor at St. Paul’s Hospital Millennium Medical College- SPHMMC, on LinkedIn, adding:
“New Cancer Hallmarks: Advancing Tumor Biology and Therapy !! Thank you Tesfaye Belete for the enlightening piece of article!!”
Quoting Tesfaye Belete‘s post:
“The Evolving Landscape of Cancer Hallmarks: Expanding Beyond the Original Six( Highly Commendable Piece from Hanahan on Cancer Hallmarks).
Recent advancements in cancer research have led to the identification of new hallmarks that enhance our understanding of tumor biology.
- Evading Immune Destruction: A core acquired capability, highlighting the complexities of the tumor microenvironment (TME), including multi-layered immunosuppression and T-cell exhaustion.
- Deregulating Cellular Metabolism: Beyond the classic “Warburg effect,” this hallmark addresses the diverse metabolic rewiring that sustains tumor growth and survival.
- Unlocking Phenotypic Plasticity: Introduced in 2022, this hallmark emphasizes the role of epigenetic flexibility, enabling tumors to adapt through state switching and heterogeneity, as well as promoting adaptive resistance.
These new hallmarks are integrated within an expanded framework that highlights hallmark-enabling characteristics, hallmark-conveying TME cells, and systemic interactions.
The Original Six Hallmarks:
1. Sustaining Proliferative Signaling.
2. Inactivating Growth Suppressors.
3. Resisting Programmed Cell Death.
4. Establishing Replicative Immortality.
5. Inducing/Accessing Vasculature.
6. Activating Invasion and Metastasis.
This ongoing evolution in our understanding of cancer hallmarks underscores the complexity and adaptability of tumors, paving the way for more targeted therapeutic approaches.
The “Hallmarks of Cancer” papers by Douglas Hanahan and Robert Weinberg are among the most highly cited in scientific literature, with the two primary, seminal papers having been cited over 100,000 times combined as of 2024-2025.”
