Miguel Zugman։ Exploring JNJ-6143 as a First-in-Class RIPTAC in Heavily Pretreated Prostate Cancer

Miguel Zugman, Medical Oncologist at Einstein Hospital Israelita, shared on X:

“First-in-class RIPTAC in mAPMR.

Thrilled to see this beautifully presented by Benjamin Garmezy at ESMO26.

JNJ-6143 is a first-in-class RIPTAC targeting AR × BRD4, designed to induce BRD4 loss of function and prostate cancer cell death.”

At 50 mg QD in heavily pretreated mCRPC:

• 69% ORR (11/16)

• 13% CR + 56% PR

• 88% DCR

• Median DOR: 11.0 months

• 77% confirmed PSA50

• 48% confirmed PSA90

PSA responses were observed across high-risk molecular features, including AR amplification, AR-V7, AR LBD mutations, TP53, PTEN and RB1 alterations.

Safety was favorable, with mostly grade 1-2 TRAEs.

At 50 mg QD, the most frequent TRAEs were nausea, headache, increased cholesterol and hypertriglyceridemia.”
