Jame Abraham: Anbenitamab Shows Promising PFS Benefit in HER2+ Advanced Breast Cancer

Jame Abraham, Enterprise Chairman and Professor at the Department of Hematology and Medical Oncology at Cleveland Clinic, shared on X:

“A new chapter in HER2+ breast cancer.

From trastuzumab to dual blockade, TKIs, and ADCs, and now bispecific antibodies.

885 patients with untreated HER2+ advanced breast cancer were randomized to anbenitamab (HER2 bispecific) + nab-docetaxel vs THP.

  • 24-mo PFS: 58.5% vs 34.4%
  • HR 0.46 (95% CI 0.37–0.56)
  • ORR: 87.6% vs 73.4%
  • Benefit across all subgroups
  • More grade ≥3 AEs (45.9% vs 34.4%), mostly neutropenia; severe diarrhea and ILD were low

OS is still immature, the trial was China-only, and the comparator was THP rather than T-DXd + pertuzumab.

Global validation is next. Most importantly, this is amazing news for patients newly diagnosed with HER2+ metastatic breast cancer.

Kudos to Prof. Binghe Xu, the Chinese investigators, and the patients who made this possible.”
