“I’m really happy to share that our work, ‘ESR1 mutations and CDK4/6 inhibitor choice shape clonal selection and adaptive cell states during acquired resistance’ has just been published!
This study represents the main project of my postdoc at Dana-Farber Cancer Institute, and I am excited to finally share our findings.
In this paper, we show that:
- ESR1 mutations are enriched in clinical samples of acquired CDK4/6i resistance and can expand to near clonality;
- ESR1 Y537S measurably reshapes clonal and epigenetic evolution under palbociclib but has a weaker impact under abemaciclib;
- Palbociclib and abemaciclib impose distinct selective pressures leading to divergent resistant states with limited clonal overlap and asymmetric cross-resistance.
