“As ADCs continue to transform the treatment landscape of metastatic breast cancer, understanding who benefits the most from these therapies is becoming increasingly important
Our latest publication in the Journal of the National Cancer Institut (Oxford University Press) addresses this question by investigating the relationship between Trop-2 expression and the efficacy of Trop-2-targeting ADCs.
In this systematic review and meta-analysis of randomized phase II–III trials, we found that:
- Trop-2 ADCs significantly improve progression-free survival compared with chemotherapy.
- The magnitude of benefit is consistently greater in Trop-2–high tumors.
- In HR+/HER2− MBC, a significant benefit was observed only in patients with high Trop-2 expression.
These findings reinforce the need for standardized and clinically meaningful Trop-2 testing to better guide patient selection and optimize treatment decisions!
