“4 years into starting the lab and we are identifying early defects in T cell surveillance that foster cancer development and designing the corresponding interception trials.
Today in Nature we present the first of these targetable axes of immune dysregulation to detect and intercept lung pre-cancer.
During early pulmonary tumorigenesis a clonally integrated eTreg network arises across
- draining lymph nodes
- blood
- preinvasive lesions
This has given us 3 tools for interception:
- A blood eTreg signature that tracks preinvasive progression and peaks in those with high risk lesions; helping to identify a population with urgent clinical need
- The early eTreg network as an actionable target
- A drug: PI3K delta inhibition dismantles the preinvasive eTreg circuit and intercepts lung cancer. The latest generation of these drugs show remarkable safety and eTreg targeting profiles.
We are seeing this axis across multiple pre-cancers – more to come on this.
This vast multidisciplinary study comes from teamwork spanning 7 co-first and 3 co-second authors across 3 labs.
