Sudha Kumari: Actin Waves Guide an Outward Movement of Microclusters in the Lymphocyte Immunological Synapse

“Thrilled to share our recent work in EMBOReports that uncovers actin waves in lymphocytes, which rescue antigen receptor loss during immune response. Super thankful to DBT/WT India Alliance, Anusandhan National Research Foundation (ANRF), Infosys, MOE-STARS.

After successful antigen recognition at the immunological synapse, some antigen receptor translocates to the cell center for downregulation, while a sizeable fraction is rescued for future engagement. The mechanism of sorting and rescue is not clear.

We used TIRF-SIM to observe the first two minutes of receptor action at the immunological synapse, and found that T cells generated outward actin waves, and used them to “kick” antigen receptors outwards, away from the downregulation zone

coupling of antigen receptor to outward waves required WASP protein, in the absence of which, although actin waves formed, most antigen receptor still ended up in the downregulation zone

While actin waves were seen in practically every primary lymphocyte type we tested, they were missing in the Jurkat T cell line. As a result, most antigen receptors ended up in the central downregulation zone here as well

Overall, we present a novel mechanism of actin-based directional sorting of surface receptors. How outward waves can co-exist with the famous centripetal flow of actin, and why primary cells have robust actin waves will be a focus of future work.
