“Excited to share the publication of our external validation of NADIR in the phase III ARCHES trial, now published in Cancer.
NADIR was developed to predict achievement of a deep PSA response (PSA ≤0.2 ng/mL at 6 months) in men with metastatic hormone-sensitive prostate cancer receiving intensified systemic therapy.
In ARCHES, the model demonstrated strong discrimination (AUC 0.80) and good calibration, providing an important independent validation across a separate phase III trial and treatment setting.
For me, the larger goal of this work is not simply prediction. We hope that early, readily available clinical information can ultimately help identify patients whose disease is responding exceptionally well—and those who may benefit from earlier treatment adaptation.
The next step is to move beyond retrospective validation toward prospective, model-guided therapeutic adaptation.
