“ESMO 2026, Day 1 highlights from Madrid
Selected findings from today’s updates:
- NATALEE: Six-year results showed a sustained benefit with adjuvant ribociclib plus endocrine therapy in eligible HR+/HER2− early breast cancer, with a 5.3 percentage-point absolute improvement in invasive disease-free survival and an encouraging overall survival signal.
- HERIZON-GEA-01: Zanidatamab plus tislelizumab and chemotherapy achieved a median overall survival of 27.0 versus 19.6 months with trastuzumab plus chemotherapy in first-line HER2-positive advanced gastroesophageal adenocarcinoma.
- G-HOPE-001: The investigational CLDN18.2-directed ADC arcotatug tavatecan improved median overall survival to 10.2 versus 6.9 months with physician’s-choice treatment in previously treated CLDN18.2-positive gastric/GEJ cancer.
- RASolute 302: Additional exploratory subgroup analyses showed a generally consistent direction of effect favouring daraxonrasib in previously treated metastatic pancreatic cancer.
- AMPLIFY-7P: The KRAS-targeted pancreatic cancer vaccine trial did not meet its primary disease-free survival endpoint. Subgroup findings provide questions for further investigation.
For me, interpreting these results means considering the absolute benefit, the population studied, and the balance between efficacy and treatment burden.”
