Muhammad Shaheer Mannan: MRD-Guided Maintenance Therapy in Newly Diagnosed Multiple Myeloma

“Honored to be a co-author on an ongoing single-arm Phase II clinical trial presented at the 2026 European Hematology Association (EHA) Congress.

MRD-Guided Maintenance Therapy with Belantamab Mafodotin and Lenalidomide After Autologous HCT in Newly Diagnosed Multiple Myeloma: Interim Analysis

This prospective Phase II study explores an important question in modern myeloma care: can measurable residual disease (MRD) be used to personalize maintenance therapy and potentially guide treatment discontinuation?

Key interim findings:

  • 56% of enrolled patients had high-risk cytogenetics
  • MRD-negative complete response by NGS improved from 18% before maintenance to 60% after one year
  • No disease progression or deaths reported during follow-up
  • Ocular adverse events were predictable, manageable, and largely limited to Grade 1–2 toxicity
  • Belantamab mafodotin administered every 12 weeks demonstrated a practical, lower-burden maintenance strategy

As the field moves toward precision-guided treatment, MRD-directed approaches may help optimize outcomes while reducing unnecessary long-term therapy exposure.

Special thanks to Qaisar Bashir for his mentorship, guidance, and support throughout my Stem Cell Transplantation and Cellular Therapy elective at The University of Texas MD Anderson Cancer Center. It was a privilege to learn from a leader in the field and contribute to research that continues to shape the future of multiple myeloma care.”
