“ASCO26 Practice-changing data in HRR-mutated mCSPC?
The phase III TALAPRO-3 trial demonstrates a significant rPFS benefit with talazoparib + enzalutamide in patients with HRR-altered mCSPC (HR 0.48; 52% reduction in the risk of progression or death).
What really caught my attention was the magnitude of benefit observed in ATM-mutated disease:
ATM: HR 0.43 (95% CI 0.24–0.79)
Historically, ATM-altered prostate cancer has been challenging, often showing less impressive responses to PARP inhibition than BRCA-altered tumors. Seeing this signal emerge in the hormone-sensitive setting is remarkable.
Could this become a new standard of care for HRR-mutated mCSPC?”
