“I’m happy to share the final version of our article ‘Mutational landscapes of brain metastases across various histological subtypes of lung cancer’, just published in Lung Cancer – the official journal of ESMO and the British Thoracic Oncology Group.
This work was supported by the SONATA grant (NCN) and is the result of a great collaboration between Uniwersytet Medyczny w Lublinie, Karolinska Institutet, Medical University of Gdansk, Wojskowy Instytut Medyczny – Państwowy Instytut Badawczy in Warsaw, and the Polish Brain Metastases Consortium.
Our work represents one of the larger sequencing-based cohorts of lung cancer brain metastases reported to date. Importantly, the cohort includes brain metastases from SCLC and LCLC, which represent exceptionally rare and valuable subgroups.
In summary, our results indicate that:
- Genetic landscape shaped by smoking habits might provide a favorable milieu for CNS colonization.
- LUAD-BM may develop through an APOBEC-driven pathway enriched for atypical, non-druggable EGFR, PTEN, and NF1 mutations, as well as a smoking-related pathway characterized by typical, potentially targetable KRAS mutations.
- SCLC-BM and LCLC-BM harbor mutations in genes associated with glial proliferation, suggesting close interaction between metastatic tumor cells and the brain microenvironment.
- High frequency of H3F3A mutations in brain metastases, together with our previous observations regarding mutations in genes regulating chromatin organization, highlights a potential role of genome architecture in lung cancer dissemination to the CNS.”
