Mali Barbi: Reprogramming the Tumor Neighborhood with Macrophage-Targeted CAR-T Cells

“Solid tumor CAR-T has been hitting a wall. The “Trojan Horse” study from Brian Brown Lab (Icahn School of Medicine at Mount Sinai) just published in Cancer Cell is a necessary shift in strategy. Instead of targeting the tumor directly, they targeted the “guards” (TAMs) using FOLR2/TREM2 CAR-Ts armored with IL-12.

The result? A total TME reset from immunosuppressive to pro-inflammatory. For aggressive diseases like Uterine Carcinosarcoma, where the TME is a fortress, we have to start targeting the “neighborhood” not just the “house.”
