“We are delighted to share our new paper in Advanced Science: ‘A Drug-Gated, Modular STAb-T Immunotherapy With External Control’.

This study introduces the first pharmacologically controllable STAb-T system, adding an external ON/OFF switch to our cell-based immunotherapy platform.

The engineered T cells secrete two inactive antibody modules that assemble into a functional T-cell engager only in the presence of a rapalog. This enables: 

  • Drug-dependent and dose-tunable T-cell activation
  • Stringent control of cytotoxic activity
  • On-demand antitumor activity in vivo
  • A modular design adaptable to different tumor antigens
  • A safety-by-design strategy to reduce uncontrolled and off-tumor activity

By combining the localized immune-recruiting capacity of STAb-T cells with reversible pharmacological control, this work provides a new framework for developing safer and more adaptable living cell therapies.

This landmark achievement for the STAb-T platform represents another important step towards the continued development and clinical translation of this technology, driven by the CNIO-HMRIB Cancer Immunotherapy Unit Spanish National Cancer Research Centre (CNIO) CNIO – Spanish National Cancer Research Centre, the Hospital del Mar Research Institute, and the Hospital 12 de Octubre, Banc de Sang i Teixits in collaboration with STAb Therapeutics.
