“I am pleased to share our latest review on Tebentafusp (Kimmtrak).
As a first-in-class ImmTAC, it was the first therapy to demonstrate a clear overall survival benefit for HLA-A*02:01–positive advanced uveal melanoma (UM) patients.
Our review arrives at a pivotal moment as the advanced UM landscape expands rapidly. While our manuscript was finalizing, key clinical milestones have continued to emerge:
The Long-Term Data: Recent 5-year survival data presented by Paul Nathan at AACR26 which just missed our publication window – demonstrates a durable survival benefit for tebentafusp over investigator’s choice (16% vs. 8%; HR: 0.67).
The Next Frontier: We now eagerly await a practice-changing update at ASCO26 from Marlana Orloff on the Phase 2/3 OptimUM-02 trial. The combination of darovasertib + crizotinib vs. ICI is expected to show a PFS advantage of 6.9 vs. 3.1 months (HR: 0.42).
This review offers a comprehensive baseline on the structural and clinical mechanics of T-cell redirection in UM as we transition into this next era of targeted combinations.
