Jinghua Hu: Breaking the Bottleneck of Pan-RAS Inhibitor Resistance

Jinghua Hu, Co-Founder at Santolecan Pharmaceutical, shared on LinkedIn:

“Breaking the Bottleneck of Pan-RAS Inhibitor Resistance: A Shift in Paradigm

The clinical reality of targeted oncology is a race against time. While next-generation pan-RAS inhibitors show remarkable initial responses, acquired resistance remains the ultimate hurdle—often emerging within ~8 months in clinical settings. High target residence time alone cannot completely block the resistance evolution of RAS-mutant tumors.

At Santolecan Pharmaceuticals, we asked a different question: Instead of trying to block a mutating target, what if we turn the tumor’s feeding habit into its own destruction?

CQ-0736’s dual-targeting therapeutic strategy shifts the paradigm completely:

  1. Exploits Macropinocytosis: Instead of attempting to block complex RAS signaling pathways, our modality leverages the hyperactivated “macropinocytosis” feeding habit of RAS-driven tumor cells, acting as a metabolic “Trojan Horse.”
  2. Targets Late-Stage Cell Growth:
It delivers precise, well-timed therapeutic intervention during critical cell cycle phases (Mitosis/M Phase), where intervening in late-stage cell growth exhibits a significantly stronger anti-resistance profile compared to targeting early stages.

With compelling head-to-head in vivo efficacy validated in an acquired RAS-inhibitor resistant model, we are advancing our IND-enabling studies.

We welcome the opportunity to share our most recent result with strategic partners and biotech investors. Please connect!”
