“Eli Lilly and Company’s olomorasib received FDA Breakthrough Therapy designation as monotherapy for adults with previously treated advanced pancreatic cancer whose tumors have a KRAS G12C mutation.
This is an important regulatory milestone in a difficult-to-treat disease where targeted options remain limited. KRAS mutations are common in pancreatic cancer, and KRAS G12C represents a much smaller molecular subset, occurring in approximately 1% to 2% of patients with pancreatic cancer.
In light of the recent Revolution Medicines approval of RASONQUE™ / daraxonrasib, this is an important distinction.
RASONQUE™ represents a broader RAS-targeted approach in metastatic pancreatic adenocarcinoma, while olomorasib is focused specifically on a rare KRAS G12C-mutant subset where no approved targeted therapy currently exists.
The designation is based on preliminary results from the Phase 1/2 LOXO-RAS-20001 study, which is evaluating olomorasib in KRAS G12C-mutant advanced solid tumors, including pancreatic cancer.
Even when a molecular subset is small, identifying an actionable alteration can clarify the development strategy and help support new treatment options for patients with historically limited choices.
As this program moves forward, I’ll be watching the maturity of the efficacy and safety data and how the development strategy evolves for this rare KRAS G12C-mutant pancreatic cancer population.”
