What we studied
In this study, we analyzed:

  • The tumor expression of B7-H4, an emerging checkpoint with growing clinical development.
  • The presence of CCR8⁺ Tregs, an immunosuppressive subpopulation enriched in the tumor microenvironment.
  • Their possible convergence as a functional immunosuppressive axis.

Our data give rise to a rational hypothesis:

  •   Anti-B7-H4 ADC → eliminates the tumor cell.
  •   CCR8 inhibitor → dismantles the intratumoral Treg niche.
  •   Result: cytotoxicity + structural immune release.

Perhaps the next frontier in immunotherapy is not to add more molecules, but to orchestrate them with mechanistic intent from early design.
