“Cytokine Storm.
Cytokine storm encompasses hyperinflammatory syndromes driven by excessive immune activation and elevated circulating cytokines, leading to tissue damage, multiorgan dysfunction, and potential lethality.
Etiologies include infection, auto-inflammatory and autoimmune diseases, monogenic disorders, and therapeutic interventions such as immunotherapy. A conserved mechanism involves aberrant cytokine release amplified by a positive feedback loop between inflammatory signaling and PANoptosis, propagating tissue injury through paracrine cell death and further cytokine production.
Defining the shared molecular pathways is essential to guide targeted modulation of inflammation, particularly in oncology where immune-based therapies can precipitate severe systemic toxicity.”
