“Advances in Basic Sciences in Thoracic Oncology and Translational Impact.
Over the past 20 years, major progress in basic and translational science has reshaped the understanding of lung cancer, mesothelioma, and thymic epithelial tumors. What was once grouped as a single disease is now recognized as a collection of biologically distinct entities with marked histologic, immune, and genomic heterogeneity, directly influencing prognosis and treatment response.
Key advances include the clinical use of immune signatures such as PD-L1 to guide immunotherapy, recognition of inherited cancer risk with an approximately 85% increased lung cancer risk among first-degree relatives, and the growing relevance of germline testing. In mesothelioma, identification of BAP1 and other germline alterations has defined less aggressive variants that benefit from tailored screening and therapy. Progress in thymic tumors has been more modest but recent biological insights are beginning to translate into clinically relevant strategies.
A clear example of how basic science has enabled precision approaches across thoracic malignancies, moving beyond uniform treatment paradigms.”
