“Adding a molecularly-targeted agent to backbone chemotherapy feels like a logical step and way to improve responses, but in this randomised study adding palbociclib to temozolomide/irinotecan did not improve outcomes for patients with Ewing sarcoma.
In fact, median EFS was worse (1.5 months) for patients on the experimental combination than those receiving standard chemotherapy (4.4 months).
Disappointing and sobering that pre-clinical data and good rationale for use of a CDK4/6 inhibitor did not translate into the clinic…”
