“Antibody-drug conjugates (ADCs) are reshaping treatment options in gynecologic oncology – and B7-H4 is emerging as one of the most promising targets.
A 2025 review published in JIPO examines B7-H4-directed agents currently in clinical development, a protein frequently overexpressed in ovarian cancer, endometrial cancer, and triple-negative breast cancer.
Key points from the review:
- B7-H4-targeted ADCs (including puxitatug samrotecan and GSK5733584) have shown early clinical activity across these tumor types
- Combination strategies pairing ADCs with checkpoint inhibitors or PARP inhibitors are showing enhanced activity in preclinical models
- Multiple trials are ongoing to define the role of these agents in ovarian and endometrial cancer specifically.”
