My notes:
- META10-19 can be manufactured from just ~100 mL of peripheral whole blood, avoiding the need for conventional leukapheresis. This simplified manufacturing process could make it accessible to patients who are too frail or otherwise unsuitable for leukapheresis.
- All patients exhibited robust in vivo expansion of META10-19, reaching a median peak of 660.7 CAR-T cells/μL, highlighting the therapy’s potent expansion kinetics and sustained persistence.
Why IL-10?
We have studied cytokines that enhance T-cell fitness for many years. While IL-10 is well known for modulating inflammatory responses through the JAK/STAT pathway, its role as a metabolic regulator has long been underappreciated. While a few recent studies suggested that IL-10 is one of the key regulators of T-cell metabolism, we had the opportunity to expand our knowledge of it’s impacts on OXPHOS and mitochondrial functions, which then lead to:
- Higher proliferation
- Less exhaustion
- More Tscm phenotype
