What gives some colorectal cancer cells the ability to spread?
We found that ceramides, a class of lipids made by cancer cells, help activate YAP and a program normally used to repair the intestine after injury.
In our experimental models, increased ceramide-regenerative signaling helped cancer cells seed metastases in the liver without substantially changing the size of the primary tumor. That suggests the ability to spread can be regulated separately from tumor growth.
A high-fat diet increased ceramide production and promoted metastasis, while reducing ceramide synthesis suppressed it.
Together, these findings help explain how dietary lipids can shape cancer cell behavior and point to new questions about how we might prevent metastatic spread.
