“ELECTRA (Phase 1b/2, interim Phase 2 data): elacestrant + abemaciclib in ER+/HER2- metastatic breast cancer with active brain metastases, presented at the 2026 SNO/ASCO CNS Metastases Conference.
Both agents cross the BBB, reaching clinically meaningful plasma and CSF concentrations.
Efficacy in the interim analysis (patients with active brain metastases, per BICR):
- Overall RECIST v1.1 (n=21): ORR 14.3%, CBR-24wk 23.8%
- Intracranial RECIST v1.1 (n=16): ORR 12.5%, CBR-24wk 25.0%
- RANO-BM (n=16): ORR 18.8%, CBR-24wk 31.3%
That’s a 2-3 times jump in ORR vs historical endocrine therapy regimens, in a population that often has limited options.
One fatal carotid thrombosis (abemaciclib-related) led to revised eligibility criteria to reduce VTE risk.
