Marco Davila: Roswell Park Advances Solid Tumor CAR T Therapy With First GB-5267 Patient

“Our team at Roswell Park Comprehensive Cancer Center has treated the first patient with GB-5267, an investigational CAR T-cell therapy for platinum-resistant ovarian cancer. This is a meaningful milestone in our collaboration with Generate:Biomedicines and our effort to extend cellular therapy to patients with solid tumors.

One of the central challenges is the tumor microenvironment, which can suppress the immune cells we engineer to attack cancer. GB-5267 brings together several approaches to address that challenge:

  • A CAR targeting MUC16, a protein expressed on many ovarian cancer cells.
  • IL-18 ‘armoring,’ designed to support immune activity within the suppressive tumor environment.
  • AI-assisted engineering aimed at improving the cells’ ability to kill cancer, expand, and persist.

The phase 1 trial, led by Emese Zsiros, is evaluating safety and tolerability. It is also testing this approach without pre-infusion lymphodepleting chemotherapy.

We are early in this work, and clinical benefit has not yet been established. Bringing a new therapy into the clinic is the beginning of learning what it can do for patients.
