High-Risk Melanoma, Important New Hope: Personalized Therapeutic mRNA Vaccine.
Problem
Melanoma is a particularly aggressive and dangerous skin cancer. Even after complete surgical removal of a high-risk melanoma, microscopic cancer cells may remain and later recurrence or metastases may occur.
The New Evolution
Early results from the Phase 3 INTerpath-001 study show that the addition of the personalized therapeutic mRNA vaccine intismeran autogene to standard immunotherapy with pembrolizumab significantly improved outcomes in patients with fully operated, high-risk melanoma.
For each patient, their tumor is analyzed and personalized mRNA therapy with selected neoantigens is created so that the immune system can better recognize cancer cells.
The Benefits
The study showed:
- Significant improvement in recurrence-free survival (RFS)
- Significant improvement in survival without distant metastases (DMFS)
- The treatment was well tolerated, with no new or unexpected side effects.
This is the first positive Phase 3 result of personalized neoantigen therapy and mRNA cancer therapy.
Where Are We Today
- The treatment remains investigational and has not yet been approved for this indication.
- The study is ongoing, among other things, to see if the benefit translates into an improvement in overall survival.
- Full analytical data are expected at an upcoming international scientific conference.
