
Jun Kim: Neuron–tumor Interactions via Axonal Injury Fuel Early Glioblastoma Progression

”Just published in Nature, Clements et al. reveal that axonal injury is a key driver of early GBM progression, overturning the assumption that early GBM growth is purely tumor-cell intrinsic and showing neurons profoundly shape GBM biology throughout their life cycle (Ciaran Scott Hill and Parrinello Labs).
While most GBM research to date has focused on advanced, therapy-resistant stages, this study highlights an unappreciated latent phase of the disease. The authors show that early tumor infiltration into the white matter physically damages axons, triggering Wallerian degeneration (WD). This process amplifies neuroinflammation and fuels tumor proliferation through a feedforward loop.
Importantly, they identify SARM1 as the central effector of WD and demonstrate that genetic inactivation of SARM1 preserves axonal integrity, suppress progression from latent to advanced disease, and improves outcomes in GBM models.”
