“Excited to share our study on immune interactions in extramedullary (lung) acute myeloid leukemia (AML) out today at Nature Immunology!
Acute respiratory failure frequently occurs in AML due to leukemia infiltration of the lungs. The question is why and what can we do to suppress it.
Varvara Paraskevopoulou in the lab started this work by mapping AML:stromal and AML:immune cell interactions using scRNA-Seq and Visium/Xenium 10x Genomics spatial tools.
We then focused on two inflammatory/immune modulating pathways, IL33:IL1RL1 and LGALS9:GAL9R. The S-type lectin Lgals9 is triggered by inflammation within infiltrated the lungs. The alarmin IL-33 and its receptor (IL-1RL1) are involved in cell–cell interactions within the leukemic lung microenvironment.
Targeting either the Lgals9 or IL-33 axis significantly decreased overall AML burden and lung infiltration through effects on both the immune microenvironment (T cells) and AML cells.
