Gilberto Lopes, Chief of the Division of Medical Oncology at the Sylvester Comprehensive Cancer Center, UICC President-Elect Candidate, shared on LnkedIn:
“5 lung cancer trials I will be watching closely at ESMO 2026.
Each addresses an important clinical decision:
- DeLLphi-305: Should tarlatamab join durvalumab in first-line maintenance for extensive-stage small cell lung cancer?
- KRAScendo 1: Can divarasib improve outcomes compared directly with sotorasib or adagrasib in previously treated KRAS G12C-mutated NSCLC?
- SAFFRON: Can osimertinib plus savolitinib improve outcomes after osimertinib progression in MET-selected EGFR-mutated NSCLC?
- OptiTROP-Lung04: Will the final overall survival analysis reinforce the published benefit of sacituzumab tirumotecan over platinum chemotherapy after EGFR-TKI progression?
- OptiTROP-Lung06: Can sacituzumab tirumotecan plus pembrolizumab improve on chemotherapy plus pembrolizumab in first-line PD-L1-negative nonsquamous NSCLC?
The premeeting signals are encouraging. Sponsor announcements report OS benefits for DeLLphi-305, KRAScendo 1 and SAFFRON. OptiTROP-Lung04 already has published PFS and interim OS benefits. For Lung06, the announcement reports a PFS benefit and a positive OS trend-not a demonstrated OS benefit.
The full presentations must establish the magnitude and durability of benefit, CNS efficacy, toxicity, quality of life and applicability to our patients.
The accompanying figure distinguishes published evidence from sponsor topline announcements, using information reviewed through October 21, 2026.
Which of these studies could most change your practice?”
