Tiago Cordeiro Felismino, Chief of Upper Gastrointestinal Oncology at A.C. Camargo Cancer Center, shared a post on LinkedIn:
“Pleased to share our meta-analysis published in the Journal of Gastrointestinal Cancer, evaluating first-line zanidatamab-based therapy in advanced HER2-positive gastroesophageal cancer.
3 trials, 685 patients
- Median OS: 26.1 months
- Median PFS: 12.6 months
- Response rate: 70%
- Grade ≥3 diarrhea: 26%
Promising efficacy, with relevant gastrointestinal toxicity.”
More thoroughly about the results of the study:
- High disease control and durable responses: Zanidatamab-based combinations achieved a pooled disease control rate of 96%, with a median duration of response of 17.1 months.
- Frequent treatment-related adverse events: Any-grade diarrhea occurred in 89% of patients, nausea in 60%, and infusion-related reactions in 30%.
- Additional safety concerns: Serious treatment-emergent adverse events occurred in 66% of patients. Immune-mediated adverse events, left ventricular dysfunction, and non-infectious pulmonary toxicity were also reported.
- Need for further validation: The findings support further evaluation of zanidatamab-based combinations, but randomized studies are needed to better establish their safety and clinical benefit.
