Including input from cellular therapy experts and key stakeholders from around the globe, this review summarizes the current landscape of point-of-care (POC) CAR T-cell manufacturing, emphasizing clinical outcomes, economic sustainability, and regulatory considerations.
- CAR T-cell therapy transformed the landscape of hematologic malignancies and is rapidly expanding into solid tumors and autoimmune diseases. Readily available products, using other platforms such as in vivo, allogeneic, and gene edited constructs are in pipeline as well.
- As of now, CAR-T’s full clinical potential remains constrained by centralized manufacturing, production costs, logistical complexity, prolonged vein-to-vein times, limited global accessibility, and regulatory frameworks that hinder rapid implementation of manufacturing innovations.
- Decentralized POC manufacturing, pioneered by academic centers, offers a promising alternative to address these limitations and expand access.”
