Angela Mastronuzzi: The current landscape of CAR T-cell therapy for CNS tumors

This review aims to explore the current landscape of CAR T-cell therapy for central nervous system (CNS) tumors, highlighting recent advancements and addressing challenges in achieving therapeutic efficacy.

Key Points:

  • Brain tumors are still associated to high mortality and morbidity and CAR T-cells could be an effective strategy to improve outcome
  • CART-cells in brain tumors possesses many challenges that need to be overcome, including immunosuppressive microenvironment, heterogeneous antigens, CAR T-cell exhaustion, cell trafficking and neurotoxicity.
  • To date there are about 40 clinical trials available to treat pediatric and adult patients affected by refractory or recurrent CNS tumors and some data have recently been published.
  • Both preclinical and clinical data have shown that locoregional delivery of CART cells is well tolerated.”

Angela Mastronuzzi is pediatric hematologist-oncologist and the Head of Neuro-Oncology Unit at Bambino Gesù Children’s Hospital (Italy). She is also a professor at the UniCamillus International Medical University, and a Board Member of AIEOP (Associazione Italiana Ematologia Oncologia Pediatrica). Her main fields of interest are CNS tumors and innovative therapies.
