“CISH – Cytokine-Inducible SH2-containing protein, an intracellular immune checkpoint in T cells.
- Part of the suppressor of cytokine signaling (SOCS) family.
- Triggered by TCR ligation or cytokines (IL-2, IL-3, GM-CSF, EPO).
Function: Inhibits T-cell activation by degrading signaling intermediates (e.g., PLC-gamma-1).
- Reduces cytokine release, proliferation, and T-cell survival.
- Operates inside the cell, unlike surface checkpoints (e.g., PD-1, CTLA-4).
- High CISH levels in TILs linked to lower reactivity to tumor neoantigens.
