3.3M people (23andMe).

  • Carrier frequency 1:15,850 (up to 1:2,000 in parts of Appalachia)
  • OR 25 overall | ~62× in never-smokers
  • No signal across 17 other cancers, no PRS interaction
  • British/Irish founder effect ~200 years ago

Strongest single germline risk factor in lung cancer so far. But rare, not the never-smoker gene.”
