”AIEOP-BFM ALL 2017 (High-Risk B-ALL)
Replacing 2 blocks of intensive chemotherapy with 2 cycles of blinatumomab significantly improved outcomes in high-risk B-ALL:
- MRD response: 76.9% vs 45.8%
- 4-year EFS: 83% vs 70.3% (HR 0.51)
- Relapse: 8.7% vs 16.2%
- CNS relapse: 0.3% vs 2.5%
- Markedly fewer infections, mucositis, pancreatitis, and allergy-related toxicities
No meaningful OS difference (93.6% vs 91%), likely reflecting highly effective blinatumomab-based salvage at relapse.
Blinatumomab delivered deeper remissions, lower relapse risk, and less toxicity without compromising survival.”
