“Our new paper in Science Advances characterizes 220 patient-derived tumor organoids (PDTOs) from 191 patients across 15 cancer types.
These 220 PDTOs are part of a larger pan-cancer collection at the Englander Institute at Weill Cornell Medicine, now over 400 PDTOs built up over the past decade.
The PDTOs are very faithful to the parent tumor. We see 93% histopathology match, 0.85 gene expression correlation, and 85% of dominant clones preserved. Expression also stays stable through ten passages.
The most interesting functional finding is around PARP inhibitors. Half the patients (58%) who would not qualify for FDA-approved PARP inhibitors had organoids that responded to talazoparib. Most carried DNA damage repair defects outside BRCA1/2. We saw strong synergy when combining talazoparib with temozolomide, especially in TP53-mutant tumors.
This has been a massive team effort. I want to thank M. Laura Martin, Hui-Hsuan (Helen) Kuo, Bhavneet Bhinder, Andrea Sboner, and Juan Miguel Mosquera for leading this paper, the Englander Institute team members who processed and assessed tissue, collected clinical data, performed profiling, analyzed data as well as our many clinical collaborators (all of them co-authors on paper).
