Isaias Hernandez: Excited to Share New Insights Into Dendritic Cells and Cancer Immunity

‘Dendritic cells control tertiary lymphoid structure development and maintenance in cancer.’

Tertiary lymphoid structures (TLSs) have emerged as one of the strongest predictors of response to cancer immunotherapy, yet the mechanisms governing their formation and maintenance have remained elusive.

In this study, we demonstrate that type 1 conventional dendritic cells (cDC1s) are the key orchestrators of functional TLSs throughout tumor development.

 Our main findings include:

  • cDC1s initiate TLS formation by priming T cells in tumor-draining lymph nodes.
  • Once TLSs are established, their persistence relies on the local accumulation of cDC1s and sustained antigen presentation to both CD4⁺ and CD8⁺ T cells.
  • These interactions preserve germinal center formation, T follicular helper cells, progenitor exhausted T cells, and antitumor IgG responses.
  • Therapeutic expansion or activation of dendritic cells enhances TLS development, opening exciting new avenues to improve cancer immunotherapy.

This work highlights cDC1s as central regulators of TLS biology and provides a strong rationale for developing dendritic cell – targeted strategies to enhance antitumor immunity.
