“Breakthrough in the Hidden Biology of Fusion Oncoproteins with AI.

Fusion oncoproteins drive many aggressive pediatric cancers, but their intrinsically disordered structures make them nearly impossible to target with traditional drug discovery methods.

A new AI model, FusOn-pLM, offers a novel approach by learning the unique properties of these elusive proteins.

Key Highlights:

  1. Fine-tuned on 44,414 fusion oncoprotein sequences – FusOn-pLM captures structural and functional features that general protein models often miss.
  2. Identifies likely resistance mutations in fusion-driven cancers, enabling novel drug resistance predictions.
  3. Outperforms leading models in predicting intrinsic disorder, puncta formation, and cellular localization, providing insights into how fusion oncoproteins drive cancer progression.
  4. Uses cosine-scheduled masked language modeling, dynamically adjusting training difficulty to improve representation quality.
