Erika Hamilton: New FDA-Approved Maintenance Regimen Targets HER2-Positive Breast Cancer

Erika Hamilton, Chief Development Officer, Late Phase and Director of the Breast Cancer Research Program, shared on LinkedIn:

“HER2CLIMB05 regimen now FDA approved for 1st line HER2 BCSM maintenance!

Amazing to now have a maintenence option for patients that have HR- tumors as well!

HER2CLIMB05 looked at HP +/- ET vs. HP +/- ET with tucatinib as maintenance after initial induction therapy with taxane HP in 1st line HER2+ bcsm.

  • PFS was lengthened by 8.6 with the addition of tucatinib
  • Benefit was regardless of HR status, but largest in in HR-
  • Study included those patients with brain mets (albeit rare in 1st line)
  • Interestingly, endocrine therapy was only used in ~50% of those with ER+ disease even though it was allowed. Those that received had much longer PFS.
