Alexander Pietras։ Study Reveals How Tumor Cells Start to Look Like Reactive Astrocytes After Radiotherapy in Glioblastoma

Alexander Pietras, Senior Lecturer/Associate Professor at Lunds universitet, shared on LinkedIn:

“Our glioblastoma recurrence atlas is now out in its final form in Cell Reports by Cell Press!

Glioblastomas almost always come back after radiotherapy, often right inside the irradiated field.

We wanted to know what the irradiated brain looks like to the tumor cells that survive.

So we followed a mouse model from healthy brain to primary tumor to post-radiotherapy recurrence, using single-cell and spatial transcriptomics.

Check out the astrocytes.

Normal astrocytes in and around the tumor become reactive, and some tumor cells start to look a lot like them, with the same mesenchymal, injury-response program.

After radiation those reactive astrocyte-like tumor cells become more common and are still there at relapse.

Part of the crosstalk between the two runs through Gas6/AXL, and the program is linked to worse survival in patients.

All the data and code are openly available, so please use them.

Huge credit to David Lindgren and Rebecca Rosberg, who led this.

Thanks also to our many collaborators from across Lund University Cancer Centre, CREATE Health – Translational Cancer Centre, and Lund Stem Cell Center in Kristian Pietras, Göran B Jönsson, Henrik Ahlenius, Johan Bengzon, and Crister Ceberg’s groups.

This was a huge team effort with vital contributions from Karolina Smoląg, Elinn Johansson, Sebastian Braun, Jonas Sjölund, Dimitra Manou, Enrico Pracucci, Bengt Phung, Katja Harbst, Pauline Jeannot, Yu Ching (Suey) So, Paulina Bolívar, PhD, Maria Malmberg.

This work was made possible by generous support from the European Research Council (ERC), Cancerfonden, Barncancerfonden, Vetenskapsrådet/Swedish Research Council, and support by Mrs. Berta Kamprad’s Cancer Foundation to the L2CancerBridge program at CREATE Health – Translational Cancer Centre.

Watch this space for more to come on reactive astrocyte targeting!”
