“Happy to see this manuscript finally in print at British Journal of Haematology. Here we investigated the role of a cell intrinsic inflammatory response mediated by dsRNA from retroelements in MPN to sAML progression. We observed that ET and PV MPNs are associated with high viral mimicry/IFN signalling and this seems to suppress progression and sAML transformation by restricting LSC expansion. sAML transformation was associated with a viral mimicry escape and LSC expansion. HMAs were able to revert this escape by re-activating a viral mimicry response in cell lines, patients samples and in vivo model, in a process that is dependent on dsRNA sensors. Altogether, we propose the ratio of IFN/LSC as a biomarker and target in MPN-sAML progression.”
