Lynch syndrome and colorectal cancer

Lynch syndrome is an inherited cancer predisposition disorder caused by pathogenic variants in genes responsible for DNA mismatch repair, including MLH1, MSH2, MSH6, PMS2, and EPCAM.

These genetic alterations impair the correction of DNA replication errors, increasing the risk of cancer development. Lynch syndrome is the most common inherited colorectal cancer syndrome and accounts for approximately 3% of newly diagnosed colorectal cancer cases. Affected individuals have a substantially increased lifetime risk of colorectal cancer, often developing it before age 50.

The syndrome is also associated with

  • endometrial
  • ovarian
  • gastric
  • other cancers.
