Circulating tumor cells (CTCs) play a crucial role in cancer progression, and in vitro CTC models are essential for studying their biological mechanisms. This study characterizes extracellular vesicles (EVs) derived from CTC lines obtained from a metastatic colorectal cancer (mCRC) patient at different stages of disease progression, particularly after therapy failure, reflecting the clonal evolution of cancer. Morphological and size analyses revealed differences among EVs from various CTC lines.
Proteomic profiling, compared with the Vesiclepedia database, showed an enrichment of proteins associated with stemness, endosomal biogenesis, and mCRC prognosis. Notably, integrin family proteins were significantly enriched in EVs from CTC lines obtained after therapy failure. The in vivo distribution of these EVs was assessed, showing significant accumulation in the liver, followed by the lungs, kidneys, and bones.
This study is among the first to highlight therapy progression-associated changes in EVs from mCRC patients using an in vitro CTC model. – Catherine Alix-Panabières.
The findings provide insights into the role of metastasis-initiating CTC-derived EVs in cancer dissemination, suggesting their relevance for studying tumor tissue distribution mechanisms. However, further validation in mCRC patients is necessary. This work underscores the potential of CTC-derived EVs as valuable tools for understanding cancer spread.”
