“Why the Blood–Brain Barrier Gets Leakier with Age.

Deficiency in N-cadherin-Akt3 signaling impairs the blood-brain barrier.

A new Cell Reports study reveals that aging disrupts the molecular ‘glue’ holding brain blood vessels together. The researchers found that N-cadherin, a key adhesion protein, stabilizes occludin at tight junctions via the PI3Kβ–Akt3 pathway.

With age—or when N-cadherin is genetically deleted—these junctions destabilize, making the blood–brain barrier more permeable and impairing short-term memory.

Understanding this pathway opens the door to new strategies for protecting brain health and preventing age-related cognitive decline.”
